Autolysis of Neisseria gonorrhoeae. Relation between mechanical stability and viability.
نویسندگان
چکیده
The relationship between the mechanical stability and the viability of N. gonorrhoeae (Type 4) in suspension was investigated. A correlation between viability and optical density recordings was often found. However, in spite of increased mechanical stability in solutions with low pH (5-2) or containing Cu++ or sucrose (10 per cent.), these environments were toxic to the gonococci. A viability preserving effect by Mg++ (4 mM), Ca++ (4 mM), spermine (0-5 mM), polyvinylpyrrolidone (10 per cent.), and low temperature (4 degrees C) was demonstrated. The possibility of improving transport media for gonococci is discussed.

Prevention of autolysis in suspensions of Neisseria gonorrhoeae by mercuric ions.
Mercuric ions were the only metal ions which prevented autolysis of Neisseria gonorrhoeae, as measured both by stabilization of optical density and by prevention of release of [3H]diaminopimelic acid.
متن کاملCell envelope of Neisseria gonorrhoeae: relationship between autolysis in buffer and the hydrolysis of peptidoglycan.
Neisseria gonorrhoeae readily underwent autolysis when suspended in N-2-hydroxyethylpiperazine-N'-2-ethanesulfonic acid (HEPES) buffer at alkaline pH values. Autolysis was inhibited by the addition of Mg2+ or other divalent cations. Autolysis was also suppressed at acid pH (pH 6.0). Suspension of cells in buffer was accompanied by the hydrolysis of peptidoglycan. The rate of peptidoglycan hydro...
متن کاملA study on the frequency of vaginal species of Mycoplasma genitalium, Gardnerella vaginalis and Neisseria gonorrhoeae among pregnant women by PCR technique
Bacterial vaginosis or non-specific vaginitis describes the disease caused by a change in the normal Flora of the vagina, which leads to the elimination of Lactobacilli, generating hydrogen peroxide and excess growth of bacteria, particularly anaerobic bacteria. This disease is the most prevalent infection of the female genital tract, and the rate of frequency of anaerobic bacteria, specificall...
متن کاملMutations affecting peptidoglycan acetylation in Neisseria gonorrhoeae and Neisseria meningitidis.
Neisseria gonorrhoeae acetylates its cell wall peptidoglycan (PG) at the C-6 position on N-acetylmuramic acid. To understand the effects of PG acetylation on PG metabolism and release of PG fragments, we have made mutations in the genes responsible for PG acetylation. An insertion mutation in a putative PG acetylase gene (designated pacA) resulted in loss of PG acetylation as detected by a high...
متن کاملNeisseria gonorrhoeae: subdivision of auxogroups by genetic transformation.
Genetic transformation was used in an attempt to subdivide the most prevalent auxotypes of Neisseria gonorrhoeae in local isolates. The large proline requiring (Pro-) group could be divided into two genetic types, as could the less common arginine requiring (Arg-) group. The large arginine, hypoxanthine, and uracil requiring (Arg- Hyp- Ura-) group could not be subdivided by this method. The gen...
